🔬 What’s new?
We used pharmacokinetic/pharmacodynamic (PKPD) modeling to explore how bacterial load influences the efficacy of the ceftazidime/avibactam + colistin combo—key agents against multidrug-resistant Klebsiella pneumoniae.

👨‍🔬 Key findings:

  • The inoculum effect (IE) significantly reduced drug efficacy at high bacterial loads (10⁸ CFU/mL)

  • CZA’s max killing effect (Emax) dropped with increasing inoculum

  • CST required higher concentrations (↑ EC50) to maintain effect

  • No synergistic modulation of the IE between drugs

💡 Takeaway:
Infections with high bacterial burden may limit the effectiveness of this antibiotic combo—highlighting the importance of integrating IE in treatment simulations.
